siRNA plasmid is another approach to express siRNA and inactivate gene function. Using DNA vector based siRNA approach, a small DNA sequence encoding a short hairpin RNA targeting the gene of interest is cloned into a commercially available vector. The insert-containing vector can be transfected into the cells, and subsequently expresses the short hairpin RNA. DICER processes the short hairpin into ~22 nt double-stranded duplexes and activates the RISC complex to cleave the mRNA target and inactivate the gene function. This service includes siRNA design, chemical synthesis, ligation, cloning, and double-stranded confirmation sequencing. At the completion of the project, you will receive an experimental report and 20 ug of siRNA construct, ready for transfection.
